Phosphate-Sensitive Kill Switch: Allows E. coli to express E2-IM2 toxin-antitoxin system sensor
This composite part allows E. coli to express the E2-IM2 toxin-antitoxin system sensitive to a phosphate-responsive repressor. This kill switch makes use of the E2-IM2 toxin-antitoxin system to make the chassis’ survival contingent on the activity of the phosphate-starvation-inducible promoter PPhoA. This dependence is established using the bacteriophage repressor cI.Under conditions of phosphate starvation, cI is expressed, shutting down the expression of the antitoxin.
